Mechanism of Proton Pumping in Complex I of the Mitochondrial Respiratory Chain

چکیده
We propose a physical mechanism of conformation-induced proton pumping in mitochondrial Complex I. The structural conformations this protein are modeled as the motion piston having positive charges on both sides. A negatively charged electron attracts piston, moving other end away from site, thereby reducing its energy and allowing to populate site. When escapes, elastic forces assist return increasing site facilitating transfer. derive Heisenberg equations for operators rewrite them form rate coupled phenomenological Langevin equation describing dynamics. This set is solved numerically. show that can be achieved within model reasonable parameters. dependencies current geometry, temperature, parameters examined.

The Assembly Pathway of Mitochondrial Respiratory Chain Complex I.
Mitochondrial complex I is the largest integral membrane enzyme of the respiratory chain and consists of 44 different subunits encoded in the mitochondrial and nuclear genome. Its biosynthesis is a highly complicated and multifaceted process involving at least 14 additional assembly factors. How these subunits assemble into a functional complex I and where the assembly factors come into play is...
متن کاملStructures and proton-pumping strategies of mitochondrial respiratory enzymes.
Enzymes of the mitochondrial respiratory chain serve as proton pumps, using the energy made available from electron transfer reactions to transport protons across the inner mitochondrial membrane and create an electrochemical gradient used for the production of ATP. The ATP synthase enzyme is reversible and can also serve as a proton pump by coupling ATP hydrolysis to proton translocation. Each...
متن کاملFunctional Dissection of the Proton Pumping Modules of Mitochondrial Complex I
Mitochondrial complex I, the largest and most complicated proton pump of the respiratory chain, links the electron transfer from NADH to ubiquinone to the pumping of four protons from the matrix into the intermembrane space. In humans, defects in complex I are involved in a wide range of degenerative disorders. Recent progress in the X-ray structural analysis of prokaryotic and eukaryotic compl...
متن کاملMitochondrial myopathies: deficiencies localized to complex I and complex III of the mitochondrial respiratory chain.
Deficiencies of the mitochondrial respiratory chain or the phosphorylation system are becoming increasingly recognized causes of myopathy and multisystem disease in man. In biochemical terms, the ones most clearly defined have been associated with impaired activities of NADH : ubiquinone reductase (Morgan-Hughes el al., 1979, 1984; Land et al., 1981; Moreadith etal., 1984), ubiquino1:cytochrome...
